月桂基硫酸鹽胰蛋白胨肉湯復蘇菌種前應準各適于該菌種生長的固體、液體培養基和培養設備，所有操作均應在符合生物安全保護及無菌條件下進行。

啟開菌種宜采用鋸開法（鋸開前用75%酒精棉消毒菌種管外壁）。用吸管將液體培養基0.3ml左右注入被啟開的菌種安瓶中并吹打，使安瓶中的凍干菌種溶解成菌懸液。
將上述菌懸液全部吸出，分別接種到固體斜面和液體培養管中，一般放置在37℃培養箱培養18~24小時。對需要特殊培養條件的細菌，如需5%~10%CO2或厭氧條件等，應分別于CO2培養箱或厭氧培養箱中培養。

這些相應的顯色底物是由產色基因和微生物部分可代謝物質組成，在特異性酶作用下，游離出產色基因顯示一定顏色，直接觀察菌落顏色即可對菌種作出鑒定。
它是一種新型分離培養基，利用顯色培養基進行微生物的篩選分離，其反應的靈敏度和特異性大大優于傳統培養基。
